Western blot was performed using H3K27me1 Recombinant Rabbit Monoclonal Antibody (Product # 703825) and a 17 kDa band corresponding to H3K27me1 was observed across the cell lines tested. Acid extracts (1% SDS) (30 µg lysate) of HeLa (Lane 1), Hep G2 (Lane 2), K-562 (Lane 3), U-2 OS (Lane 4), PC-3 (Lane 5), T-47D (Lane 6), A549 (Lane 7) and NIH/3T3 (Lane 8) were electrophoresed using Novex® NuPAGE® 10 % Bis-Tris gel (Product # NP0301BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:500 dilution) and detected by chemiluminescence with Goat Anti-Rabbit IgG (Heavy Chain) Superclonal™ Secondary Antibody, HRP (Product # A27036, 1:5000 dilution) using the iBright FL 1500 (Product # A44115). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

For immunofluorescence analysis, HeLa cells were fixed and permeabilized with 0.5% Triton X-100 for detection of endogenous H3K27me1 using H3K27me1 Recombinant Rabbit Monoclonal Antibody (Product # 703825, 1:100 dilution) and labeled with Donkey anti-Rabbit IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor Plus 488 (Product # A32790, 1:2000). Panel a) shows representative cells that were stained for detection and localization of H3K27me1 protein (green), Panel b) is stained for nuclei (blue) using ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). Panel c) represents cytoskeletal F-actin staining using Rhodamine Phalloidin (Product # R415, 1:500). Panel d) is a composite image of Panels a, b and c clearly demonstrating nuclear localization of H3K27me1. Panel e) represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.
